Styrene and Styrene Oxide Induced Sister Chromatid Exchanges in the Cultured Human Whole Blood Lymphocytes


Abstract
This study was designed to investigate the effect of styrene and styrene-7,8-oxide(styrene oxide) on their potential to induce of sister chromatid exchanges(SC&) in phytohaemagglutinin-stimulated human whole blood lymphocytes cultured for 72 hours. The treatment with styrene or styrene oxide diluted by acetone were carried out 48 hour before harvest. The concentration of styrene was 1,0, 1.5 and 2.0 mM and that of styrene oxide was 50 and 100 M, and control group was treated with acetone only.
A dose-dependent increase of SCEs was observed in lymphocytes treated with styrene or with styrene oxide. The toxicity of styrene oxide was about 15 times of that of styrene cone sidering the ability of inducing SCE, in the cultured human lymphocytes and cell replication index. This study result was on the line that the active metabolite formed from styrene through the action of the erythrocytes is styrene oxide.
